The best org'n to join depends where you are. The PTUA is the biggest
in Australia but is mainly Victorian-based (we do have a small branch
in Perth and a bigger one in Geelong).

In Adelaide theres PPT - people for public transport - which is (or
appears to be) a fairly large and effective organisation.

Sydney has Action for Public Transport but theyre a lot smaller than
PTUA. I don't know of a group of any reasonable size in Queensland
though someone was trying to set up a PTUA up there.
